The integrated genome map of Mycobacterium leprae.

چکیده
The integrated map of the Mycobacterium leprae genome unveiled for the first time the genomic organization of this obligate intracellular parasite. Selected cosmid clones, isolated from a genomic library created in the cosmid vector Lorist6, were identified as representing nearly the complete genome and were subsequently used in the M. leprae genome sequencing project. Now a new version of the integrated map of M. leprae can be presented, combining the mapping results from the Lorist6 cosmids with data obtained from a second genomic library constructed in an Escherichia coli-mycobacterium shuttle cosmid, pYUB18. More than 98% of the M. leprae genome is now covered by overlapping large insert genomic clones representing a renewable source of well defined DNA segments and a powerful tool for functional genomics.

A RAPID AND SIMPLE METHOD OF DNA EXTRACTION FROM CLINICAL SPECIMENS CONTAINING MYCOBACTERIUM LEPRAE FOR PCR ASSAYS
We describe a rapid, simple, and reliable procedure for routine isolation of Mycobacterium leprae DNA from slit-skin swab specimens. This one-step DNA extraction method is based on the utilization of ChelexR 100, a chelating ion exchange resin. Slit-skin swab specimens from seven untreated leprosy patients at Baba Baghi Leprosy Hospital in Iran were processed by this procedure. The polymer...

The nucleotide sequence of 1.5 Mb of genomic DNA from Mycobacterium leprae was determined using computer-assisted multiplex sequencing technology. This brings the 2.8-Mb M. leprae genome sequence to approximately 66% completion. The sequences, derived from 43 recombinant cosmids, contain 1046 putative protein-coding genes, 44 repetitive regions, 3 tRNAs, and 15 tRNAs. The gene density of one pe...
متن کاملComputational genome analyses of metabolic enzymes in Mycobacterium leprae for drug target identification
Leprosy is an infectious disease caused by Mycobacterium leprae. M. leprae has undergone a major reductive evolution leaving a minimal set of functional genes for survival. It remains non-cultivable. As M. leprae develops resistance against most of the drugs, novel drug targets are required in order to design new drugs. As most of the essential genes mediate several biosynthetic and metabolic p...

متن کاملReconstructing the ancestor of Mycobacterium leprae: the dynamics of gene loss and genome reduction.
We have reconstructed the gene content and order of the last common ancestor of the human pathogens Mycobacterium leprae and Mycobacterium tuberculosis. During the reductive evolution of M. leprae, 1537 of 2977 ancestral genes were lost, among which we found 177 previously unnoticed pseudogenes. We find evidence that a massive gene inactivation took place very recently in the M. leprae lineage,...
